Learn to program the remote to your 2010 Toyota Highlander without any help from the dealer. This includes the XLS model.
Programming Instructions: Note: This is the same for many Toyota vehicles.
Remove the key from the ignition. Lock all of the doors, but open the driver’s side door. Leave it unlocked. Insert the key into the ignition twice within five seconds. Don’t turn it. Close then open the door twice. Insert the key again then remove it from the ignition.
Close then open the driver’s side door. Close then open the driver’s side door again. Insert the key. Leave it in the ignition. Close the door. Turn the key to “On,” without starting the engine, and then turn it off. Remove the key. You’ll hear the door locks sound.
Push and hold “Lock” and “Unlock” buttons on the remote for two seconds, and then release the buttons. Then push and hold “Lock” for three seconds. The remote is now programmed.

I tried this programming sequence many times on my Toyota Highlander 2010 (assembled in Nov 2009 in USA) and I don’t hear the door locks sound to confirm that the car is in the programming mode. It works great on my Camry 2005 though.
Thanks for your help

I have tried to program a key fob on 2008 and 2010 highlander (base model) and unable to get to program mode. It works if your vehicle is made before 2008. The instruction that is given on this website works for earlier model. You have to take to a dealer to have it programmed because you need a scan tool. If anyone is successful programming it without a scan tool please let us know. Please don’t waste your time trying it.
